  • Gujarat has reported 35 laboratory-confirmed cases and 22 child deaths during the current monsoon season, while Rajasthan has also confirmed infections in border districts, prompting intensified surveillance and scientific investigations.

  • Chandipura Virus (CHPV) is an arbovirus belonging to the Rhabdoviridae family under the Vesiculovirus genus.
  • It was first identified in 1965 from Chandipura village in Maharashtra.
  • The disease primarily affects children below 15 years of age and is associated with Acute Encephalitis Syndrome (AES).
  • CHPV is transmitted mainly through the bite of infected sandflies (Phlebotomus species), while the role of mosquitoes and other arthropods is under investigation.

  • Concurrently, global health bodies like the USFDA and Australia’s Therapeutic Goods Administration (TGA) have enforced rigorous, updated Acceptable Intake (AI) limits and compliance timelines for global drug manufacturers to eliminate these genotoxic impurities.
What are Nitrosamines?
  • Nitrosamines (formally N-nitrosamines) are a class of organic chemical compounds characterized by the Rβ‚‚N-N=O functional group, where a nitroso group (NO⁺) is directly bonded to a deprotonated amine.
  • They are classified as "probable human carcinogens" and are designated as a Cohort of Concern under the International Council for Harmonisation (ICH) M7 guidelines due to their exceptional mutagenic potency.
Mechanisms of Formation

Nitrosamines are not intentionally added but are formed as secondary byproducts when amines (secondary or tertiary) chemically react with nitrites or nitrates under specific conditions, including:
  • Acidic environments (such as gastric juices in the human stomach).
  • High thermal exposure (such as frying or grilling food).
  • The presence of oxidising agents or contaminated solvents.
Primary Sources of Human Exposure
  • Dietary Sources: Found in cured, fried, or processed meats (due to sodium nitrite preservatives), processed fish, beer, and fermented foods.
  • Drinking Water: N-nitrosodimethylamine (NDMA) is a highly potent nitrosamine that forms during water chlorination/chloramine disinfection when chlorine reacts with local organic nitrogen compounds.
  • Pharmaceuticals (NDSRIs): Nitrosamine Drug Substance-Related Impurities (NDSRIs) form during manufacturing scale-up, or when active pharmaceutical ingredients (APIs) interact with residual nitrites in excipients or packaging material under heat and moisture.
  • Tobacco: Tobacco-specific nitrosamines (TSNAs) are major components in cigarette smoke.
Why are Nitrosamines Excessively Dangerous?
  • DNA Adduct Formation: Inside the human body, nitrosamines are metabolically activated by Cytochrome P450 enzymes. This activation leads to the formation of reactive ions that damage DNA by binding to it (forming DNA adducts), triggering mutations that progress into stomach, colorectal, liver, or pancreatic cancers.
  • No "Safe" Threshold: Unlike typical chemical impurities, genotoxic impurities can cause damage even at nanogram (ng) levels over long periods of cumulative exposure.

 MSME Sustainable ZED Certification Scheme
